Disorders of the nervous system follow a sequence of primary and secondary pathophysiological events, which lead from a primary cellular defect to the disease phenotype. The analysis of these sequences requires complex models. Specific gene knockout in mice, generation of gene mutants in drosophila and other organisms have provided powerful tools for analysis of disease mechanisms. Moreover, the establishment of organotypic cell culture models is an excellent basis for analysis of regulatory genes in a functional context. These models do not only allow the analysis of the consequences of gene deletion or overexpression, but also the characterization of secondary events and their role in the pathogenesis of neurological and psychiatric disorders. The collaborative research centre 581 has been designed to provide insights into the sequence of disease processes, on the basis of research on function of signaling molecules in neurons and glial cells. This interdisciplinary approach involves groups in the Institutes of Anatomy, Cell Biology, Genetics and Immunology, but also clinical research groups from the Institutes for Neurology and Psychiatry. On a long-term perspective, a better understanding of complex disease processes should lead to new therapeutic approaches for these disorders.
