Project C6 uses the immunological concept of enforced viral replication to enhance vaccine responses against chronic viruses. The PI Lang showed that CD169+ macrophages replicate virus to allow antigen presentation and efficient priming of T cell responses. He defined several molecules that are critically involved in the mechanism of enforced virus replication. The molecular targets will now be blocked or induced with existing antibodies to improve enforced viral replication during therapeutic vaccination against chronic infections. The novel strategy will be tested in three animal models, VSV-HIV infection, chronic FV infection and HIV infection of humanized BLT mice.
