The recongition of multivalent peptidic sequences by tandemly arranged adaptor domains is of importance for many intracellular signaling events. The inhibition of these kind of interactions by optimized bivalent ligands is the focus of project C4. Based on the tandem-WW domains of the spliceosomal protein FBP21 and the tandem-SH2 domains of the T cell kinase ZAP70 proline analog containing or phosphotyrosine mimetic ligands will be developed, respectively. These ligands are then targeted to the inside of the cell by nanocarriers in order to analyse their mode of action and to functionally measure their inhibitory potential.
