Functional dynamics and molecular modelling of the promyelocytic leukemia (PML) nuclear body

Promyelocytic leukemia nuclear bodies (PML NBs) are nuclearmultiprotein complexes in mammallian cells that have been linkedto a variety of cellular functions, including transcriptionalregulation, tumor suppression, cellular senescence, apoptosis,genome stabilty and anti-viral activity. The precise biochemicalrole of PML NBs in these processes, however, remainselusive. Our aim is to gain more insight into the biochemicalfunction, formation, and dynamics of these subnuclear structuresand its component parts. By applying a combination of noveloptical analysis methods, such as fluorescence recovery afterphotobleaching (FRAP), fluorescence correlation spectroscopy(FCS), fluorescence resonance energy transfer (FRET), fluorescencecross correlation (FCCS) and continuous fluorescencephotobleaching (CP) in living cells we will determine the fullrange of intranuclear mobilities and interactions of PML NBcomponentes. From these data a computer-based kinetic frameworkfor the quantitative description of the dynamics of PML NBswill be established. Our results will contribute to our understandingof the dynamic structure, function and biochemistry ofthe PML nuclear body and its component parts.
