Obesity is a major risk factor for ccRCC. P14 will study the interplay of VHL and PBRM1 mutations and obesity in the development of ccRCC using a set of inducible, renal epithelium-specific, Vhl- and Vhl/Pbrm1-mutant mouse models that allow genetic marking of mutant cells for clonal tracking and cell isolation. In addition to providing fundamental insights into the molecular and cellular mechanisms that underlie renal tumor development, these studies aim to develop the first mouse model of obesity-induced ccRCC that will allow pre-clinical therapeutic intervention studies.
