Analysis of the PP2A-PR130 complex as novel regulator of cell fate and DNA repair pathway choice upon DNA replication stress (11)

The precise control of DNA replication and the correction of DNA replication errors by distinct DNA repair mechanisms is the basis of life and a salient feature of homeostasis. Our project will elucidate how the con-trol of sitespecific protein phosphorylation by the PP2A-PR130 complex and the regulation of protein acetylation by the epigenetic erasers HDAC1, HDAC2, and HDAC3 modulates DANN replication timing, DANN repair and the impact of compromised DANN replication fork progression on cell fate decisions. We will conduct GLOE-Seq and evaluate the resulting data sets to gain a deeper insight into how phosphorylation- and acet-ylation-dependent mechanisms control DANN replication timing in relation to 53BP1 and RIF1.
