This project aims at elucidating how DNA lesions are processed during genome replication. We have devel-oped a method for the high-resolution mapping of DNA singlestrand breaks and base lesions and will now implement additional features to this tool to enhance its versatility. In combination with a set of affinity probes selective for the pathway of ubiquitindependent DANN damage tolerance, this approach will serve to detect relevant regions of the genome where DANN damage bypass is important and to identify critical factors that provide information about the mechanism of damage processing.
