We are investigating the interplay between DNA repair and epigenetics using a stabilised, chemically ma-nipulated version of the drug Decitabine (azadC). We will insert the compound into oligonucleotides for bio-chemical and biophysical studies. Structural analyses by X-ray crystallography in complex with repair pro-teins and lesion tolerance polymerases as well as detailed repair studies will be performed. Finally, we plan to use a systems approach with a newly developed direct sequencing method to learn about the positions where epigenetic changes occur in the genome due to repair and/or blocking of Dnmt enzymes.
