G-protein-coupled receptors (GPCRs) mediate the intracellular effects of a multitude of systemic and local mediators and thereby importantly modulate endothelial function. To define new roles for GPCRs in endothelial cells (EC), we performed a single-cell GPCR expression analysis in freshly isolated murine EC from different vascular beds and disease states. We identified a number of GPCRs selectively expressed in inflamed EC from brain, aorta and lung. Focusing on three orphan GPCRs that have not yet been implicated in endothelial biology, we here will analyze these orphan receptors in vitro and in tamoxifen-inducible, EC-specific knockout mice in vivo.
