For a selective GvL effect without GvHD promising targets of CD4+ T cells are DM-sensitive antigens, whose immunogenicity has already been confirmed in a mouse model in the first period. To analyze clinical relevance, the frequency of allogeneic T cells directed against DM-sensitive antigens will be correlated with the GvL effect and GvHD after HSCT. The influence of 5-azacytidine on the regulation of HLA-DO has already been observed in vitro and will now be further investigated in primary leukemia cells. Finally, T cells directed against DM-sensitive antigens in HLA-DP mismatched HSCT will be isolated, sequenced and TCR transgenic T cells will be explored for potential clinical application.
