We focused on the identification of protein modifications and interactions of key components in insulin/IGF and target-of-rapamycin (mTORC) signaling. The serum-and-glucocorticoid-inducible kinase SGK-1 couples both pathways via its dynamic localization and extensive interactions and modifications of its key components. Here, we propose genetic and protein-biochemical methods in C. elegans and cell culture models to understand how stress and nutrition modulate the feedback regulation of SGK-1 via the FOXO transcription factor DAF-16 and the mTORC2 interactor Rictor.
