Myo-inositol and its role in metabolic regulation of two contrasting high-yielding laying hen strains

Myo-inositol (MI) is the end product of complete dephosphorylation of phytate by endogenous phytases and phosphatases in the gastrointestinal tract of laying hens with an efficient phosphorus utilization. MI is known to be a component of cell membranes and of intracellular phosphoinositide- and inositolphosphate-dependent signaling pathways. Thereby, it is an important key molecule in healthy function of body cells. However, its role in laying hens is not well known yet. By a metabolomics approach, as a major finding, we detected a significant positive association of concentrations of MI with serotonin and dopamine in plasma of broilers. The aim of this study is to assess the mechanisms of absorption of MI, its metabolism and role for systemic metabolomics. Expression of intestinal MI transporters and of key enzymes in body-own metabolism of MI will be studied in laying hens of the Research Unit. Samples will be taken from the hens used in the experiments described in project 1. Experimental variables include the concentrations of calcium and phosphorus in the diet, the laying hen genotype, and the age of the hens. Plasma of all hens will be used to perform a metabolomics analysis to assess new metabolic features of MI-dependent pathways. In tissues, key enzymes of MI- metabolism will be semiquantitatively measured on protein level, to assess their existence and extent of expression. These data should serve as a basis for further functional characterization of MI metabolism on laying hens. In context with the data of all other projects, a holistic analysis of laying hen metabolism will be performed to identify animals with efficient P utilization.
