In this work package, we will describe the role of meningeal lymphatic vessels (MLV) in the physiological immune surveillance of the meningeal spaces. We will address the role of DCs associated with MLV in ensuring tolerance to CNS-derived self-antigens and their role in initiation, progression and severity of EAE. We will characterize the human MLV and study their role in maintaining the CNS immune-privilege status exerts T cell tolerance. Together, these studies will provide new insights on the role of meningeal DC in limiting or enhancing CNS autoimmune responses.
