Die Rolle der NADPH-Oxidase in der Interaktion zwischen Pflanze und Nematoden

Zusammenfassung der Projektergebnisse

The production of Reactive Oxygen species (ROS) is an important component of plant defence mechanisms. The major source of ROS during an oxidative burst in plants is plasma membranebound respiratory burst oxidase homolog (Rboh). There are ten Rboh genes (RbohA-RbohJ) in Arabidopsis. Heterodera schachtii is a cyst nematode that establishes a long-term biotrophic relationship with the host plants, including Arabidopsis thaliana. Infective stage juveniles of nematodes (J2s) invade the roots and move towards the vascular cylinder where they establish a syncytial nurse cell system. Syncytium is the only source of nutrients for nematodes throughout their life-span of several weeks. Previous loss-of-function mutants for Arabidopsis Rboh genes found that the number of infection of cyst nematodes decreased greatly in rbohD and rbohD/F compared with Col-0 plants. Here, using detailed genetic and biochemical analyses, we identified WALLS ARE THIN1 (WAT1), an auxin transporter, as a downstream target of Rboh-mediated ROS during cyst nematode infections. We found that expression of WAT1 is induced upon cyst nematode infection in RbohD/F-dependent manner and this activation is required to modulate the plant’s indole metabolism, including IAA levels, in infected cells. We also identified ROS motifs in WAT1 promoter that are required for activation of its expression by Rboh-mediated ROS. Our genetic analysis found that reprogramming of WAT1-mediated indole reprogramming is necessary for successful establishment of the cyst nematodes. In conclusion, this work clarifies a unique mechanism that enables cyst nematodes to use the host’s ROS for their own benefit.
