TRPA1 modulation by its lipid environment and physiological functions in skin-derived cells

TRPA1 is predominantly expressed in sensory neurones of the dorsal root and trigeminal ganglia and is a central molecule for sensing chemical and physical stimuli. In the proposed project, the first focus lies on the modification of the biophysical properties of TRPA1 by its lipid environment. As TRPA1 channels are physiologically expressed in highly bended free nerve endings, we will examine how inducing membrane curvature, varying the composition and surface charge of the lipid membrane and the acute addition of phospholipids affect TRPA1 gating. The second focus lies on clarifying physiological roles of TRPA1 in skin-derived cells, including a possible up-regulation on TRPA1 in malignant melanoma cells and a possible contribution of TRPA1 to painful sensations arising during photodynamic therapy.
