Endothelial responses to blood-borne mediators or mechanical stimuli are important determinants of vascular function. In the last funding period, we identified a signalling mechanism involving the cation channel Piezo1 and the Gq/G11-mediated signalling pathway, which plays an important role in endothelial sensing of laminar flow. The aim of the new funding period is to further analyse the composition of the upstream mechanism of endothelial flow sensing and the molecular mechanism of mechanotransduction linking initial mechanosensitive processes induced by laminar and disturbed flow with downstream effector functions in endothelial cells under normal and diseased conditions.
