Previously we have demonstrated that different vesicle populations of Xenopus egg extract are required for nuclear envelope assembly. One type of vesicles binds to chromatin where they fuse and form a double layered nuclear membrane lacking pore complexes. The other vesicle population binds to the preexisting double membrane and induces nuclear pore complex formation. The aim of the proposed project is to characterize the biochemical composition of the pore complex inducing membrane vesicles by mass spectrometry, gel electrophoresis and immunoblotting experiments in order to identify factors that initiate the insertion of pore complexes into the nuclear membrane. In addition we have shown that the vesicles required for nuclear pore complex assembly are transported along microtubules to the pore-free nuclei, probably by a kinesin motor protein. We plan to identify the components involved in this specific transport process. Finally, we will analyze dynamic events of vesicle fusions and pore complex formation. Different vesicle populations will be labelled with different lipophilic membrane dyes or antibodies to marker proteins and the order of events in the process of recruitment of the vesicles to chromatin will be analyzed by confocal laser scanning fluorescence microscopy.
