Neuronal Polarization and Axon Formation in Mice deficient in Rac1

The actin cytoskeleton plays an important role in neuronal polarization. Rho GTPases are major regulators of the cytoskeleton and have also been implicated in the regulation of neuronal morphogenesis. We will analyze changes in neuronal polarization using mice that are deficient in Rac1 both in vivo and in vitro. Specifically, we will perform a phenotypic characterization of the brains of Nes-Cr +/- Rac1fl/fl mice focussing on axonal pathways and dendritic layers. We will culture hippocampal neurons and cerebellar granule neurons from Rac1-knockout mice and examine axonal/dendritic number, growth rate, the actin cytoskeleton and growth cone dynamics. Furthermore, we analyze the activation and localization of Rac-effector molecules which we subsequently will manipulate. Together, this project will help to better understand the physiological role of Rac1 and their effectors in neuronal polarization.
