Final Report Abstract

The aim of WP 2.4 was to immobilize polysialic acid on the surfaces of biomaterials in order to provide materials for the construction of scaffolds for biomedical purposes, especially nerve regeneration. WP 2.4 succeeded in synthesizing nanoporous silica nanoparticles (NPSNPs) and in their functionalization, most importantly with polySia itself, but also with a growth factor and with fluorescent entities (fluorescent nanoparticles, fluorescent dyes) for imaging. Cytocompatibility testing accompanied the preparative work. WP 2.4 also succeeded in the fabrication of functionalized collagen tubes to be used as nerve guidance channels. These consisted of rolled collagen sheets decorated with NPSNPs carrying polySia. Regarding the high degrees of functionalization, both of the nanoparticles with polySia and of the collagen sheets with nanoparticles, it can be presumed that these constructs exhibit only polySia strands on their inner surface. They should thus offer a favourable milieu for nerve regeneration. As all kinds of tests performed on these scaffolds (cytocompatibility, mechanical and chemical stability) gave positive results, these are currently being tested as nerve guidance channels. In addition, it was possible to obtain semicrystalline polySia. With structural modelling, it was possible to propose a satisfactory, but not yet sufficiently substantiated model for the structure of crystalline polySia. In the course of studies, WP 2.4 interacted with nearly all the other work packages.
