Protein translocation across membranes is central for cellular function and involves specific membrane integral proteinaceous complexes. The translocon of the outer plastidic membrane (TOC) is composed of at least four subunits - the three receptor proteins Toc34, Toc64 and Toc159, and the barrel shaped channel Toc75. While the global function of the TOC is described, not much is known about the mechanisms at molecular and atomistic levels. We will move toward a structural analysis of the entire TOC complex in addition to the characterization of the GTPase cycle of Toc34 and Toc159 as central part for the translocation. This information will be unified with the exploration of the dynamics of this complex in different developmental stages and tissues to provide a basis for a molecular understanding of the translocation process in general.
