ABCB9 forms a homodimeric complex, which can functionally be divided into the core complex and the N-terminal, transmembrane domainTMD0, responsible for lysosomal localization and interaction with LAMP-1/2. We will determine the molecular structure by NMR spectroscopy. Further, the interface between TMD0 and its interaction partners will be investigated. With cell biological methods the machinery for lysosomal trafficking of ABCB9 will be studied. Peptide binding and release as well as ATP hydrolysis will be analyzed by single molecule techniques and fast kinetics.
