Cortico-spinal modulation of somatosensation

This research project aims to investigate the role of cortical layer 5 (L5) pathways in modulating the flow of sensory signals within the central nervous system, with a particular focus on the potential of L5 to suppress sensory gain and sensory-evoked behavior. This investigation is predicated on recent findings from our laboratory, wherein we discovered that L5 neurons in the primary somatosensory cortex (S1) can reduce sensitivity to somatosensory stimuli applied to the hind-paw of mice. Our aim is to explore the underlying circuits. Based on our preliminary results of the L5 projection targets, the study will concentrate on the L5 downstream circuits that are vital for sensory gain control functions, specifically those involving the dorsal horn (DH) and the periaqueductal gray (PAG). We plan to utilize virus-mediated methods, optogenetic and chemogenetic tools, electrophysiological recordings, and behavioral readouts. The project intends to anatomically characterize S1-recipient neurons in the DH and PAG, evaluate their role in sensory-driven behavior, and functionally and molecularly characterize the cortico-spinal-thalamic circuitry.
