NAADP is the most potent Ca2+ releasing second messenger known today. Among many other cell types, NAADP also operates in neurons. We hypothesize that NAADP signalling is required for oscillatory Ca2+ signalling and survival during physiological glutamate stimulation, but contributes to neuronal Ca2+ overload and cell death during excitotoxic glutamate stimulation. We will analyse how NAADP receptors and NAADP generating enzymes shape NAADP evoked Ca2+ signalling in neurons, modulate neuroprotection vs neurodegeneration in vitro as well as in mouse models for multiple sclerosis in vivo.
