Mitochondria are major sites of cellular lipid biosynthesis. The mitochondrial signature lipid cardiolipin is synthesized in the mitochondrial inner membrane. This process depends on the import of phosphatidic acid, which is transported from the mitochondrial outer to the inner membrane by the lipid transfer protein complex Ups1/Mdm35. In this project we will decipher how the physico-chemical characteristics of the membrane and the oligomeric state of the protein complex, influence protein-membrane interaction, allow the extraction and insertion of a lipid from and into membranes and determine the specific direction of lipid transfer.
