B05 investigates two orthologous proteins from Arabidopsis and wheat, studying functional differences within a species and between orthologous proteins. RADICAL-INDUCED CELL DEATH 1 (AtRCD1) andits wheat ortholog influence plant tolerance to oxidative stress and/or salinity. Analyses focus on nsSNPs in the essential WWE-PARP domain, crucial for oligomer formation and binding to stress-induced poly(ADP-ribose) chains. Methodologies compare proteoforms biochemically and structurally (x-ray crystallography), revealing differences in WWE and PARP domain cooperation and their impact on oxidative stress tolerance.
