Project A02 aims to understand the impact of allelic variation on DNA-binding and dimerization in the bHLH transcription factor PIF4 and its regulator ELF3, both crucial for plant responses to temperature changes. The project employs DNA-binding assays, structural approaches (XL-MS), and functional analyses of expressed proteoforms in Arabidopsis. Image-based phenotyping will quantify growth patterns and architectural effects, focusing on light, temperature responses, and the circadian clock regulated by PIF4 and ELF3. This project will furthermore establish in vivo XL-MS in plants.
