Expanded hexanucleotide (GGGGCC)n repeats in the C9orf72 gene account for 30% of familial ALS cases. These repeats result in the accumulation of abnormal nuclear foci and cytosolic stress granules (SGs) in motor neurons. In this project, we will conduct a comprehensive system-wide analysis to define converging alterations in interconnected RNA and protein fidelity processes induced by mutant C9orf72-associated nuclear RNA foci and cytosolic SGs.
