Recently, we have shown that an age-associated increase in average RNAPII elongation speed impairs the production of mRNA. However, we lack understanding about the molecular factors contributing to specific splicing changes and other mRNA sequence errors. We will create cellular models of accelerated and slowed-down RNAPII and profile those models by separately analyzing their nuclear and cytoplasmic transcriptomes. We will investigate the consequences of speed changes for transcript structures and protein levels in cellular models, mouse models and human kidney samples.
