The requested liquid chromatography mass spectrometry (LC-MS) system is essential for the implementation of latest MS-based methods for high-end multifaceted proteomics research at the University of Würzburg. The MS system offers highly effective, complementary peptide fragmentation methods for the identification and quantification of peptides/proteins, precise site localization of posttranslational modifications in proteins or the detection of low abundant cross-linked peptides. Through the combination of different mass analyzers, the instrument features MSn and advanced multistage acquisition modes at high mass resolution, high sensitivity, and high scan speed. Moreover, its high-mass HMRn+ option enables the analysis of intact proteins and protein complexes at high resolution. The system is highly suitable for quantitative proteomics studies, employing label-free approaches, stable isotope labelling, or isobaric mass tagging technology. In addition, the instrument provides maximum flexibility in acquisition modes for high-end LC-MS analyses, with data-dependent acquisition enhanced by advanced peak detection, data-independent acquisition, or parallel reaction monitoring for the accurate quantification of target peptide ions. These MS technologies will enable and propel current and future proteomics research projects in Würzburg, with the overarching aim to expand current knowledge about the complex organization, regulation, and dynamics of the cellular proteome in health and disease.
