The aim of this study is to develop alternative approaches for cell replacement therapies and examines the molecular mechanisms that regulate neural progenitor cell fate during development. Specifically, in vivo and in vitro approaches to study transient Notch1 signaling will be carried out to test the hypothesis that progenitors expressing transiently Notch have broad developmental potentials in vivo. Fate mapping and transplantation of cells that have transiently expressed an activated form of the Notch1 receptor (ActN) will be done using a novel conditionally-active retroviral vector approach developed in the Fishell laboratory. These cells will be analyzed in vivo, in vitro and after transplantation in disease and injury models.
