The goal of this proposal is to combine to complementary genetic technologies in order to identify key genes that control production of new neurons from neuronal stem cells in the adult brain. Based on current work on quantitative trait loci analysis (QTL) in recombinant inbred (RI) strains of mice we will use recombinant inbred intercrosses (RIX) to refine gene loci with high correlation to stem cell division and adult neurogenesis. In a parallel effort we will microdissect tissue samples from the hippocampus of selected substrains of the previously investigated recombinant inbred mice (RI), obtain cDNA from these samples and examine gene expression profiles using Affymetrix arrays under normal and under conditions known to stimulate adult neurogenesis. Via existing data bases gene array data can be related to identified key gene loci and will thus receive a context, in which they can be interpreted. Thus the power and efficiency of our gene search approach are greatly enhanced.
