Defining the regulation and role of bone marrow adipocytes during inflammatory bone loss (B01)

Project B01 aims to investigate how inflammation leads to the depletion of BMAd, and how these changes affect inflammatory bone loss, i.e. the crosstalk with inflammatory and bone cells. To that end, B01 will investigate the interactions of BMAd with osteoblasts, which are also reduced during inflammation, and osteoclasts and neutrophils, which expand during inflammatory arthritis, using co-cultures and mouse models. Moreover, B01 will assess how BMAd adapt their metabolic program to inflammatory cues (such as IL-17) using comprehensive metabolomics analyses. The long-term goal of B01 project is to understand how various cell types compete for nutrients in the bone marrow microenvironment under chronic inflammatory conditions and how depriving bone cells of nutrients contributes to bone loss.
