Two pore channels (TPCs) are endo-lysosomal cation channels that belong to the TRP channel family. The proposal builds on preliminary work showing that TPC2s promote cardiac hypertrophy in mice and that their targeted deletion reduces disease severity. Interestingly, deletion of TPC2 promotes the release of exosomes from mouse cardiac fibroblasts. Moreover, exosomes from wild-type or TPC2-/- cardiac fibroblasts appear to contain increased levels of miRNAs that promote (WT exosomes) or prevent (TPC2-/- exosomes) cardiac hypertrophy/fibrosis. In three aims we will examine the hypothesis that TPC2s and exosomes individually and in concert regulate function in normal and diseased cardiac cells and tissues.
