Post-translational regulation of signaling complexes by ubiquitylation and deubiquitylation critically contributes to the outcome of death receptor activation. This project will investigate how the protein SPATA2 regulates cell death decision-making processes by recruiting the deubiquitinase CYLD to the TNF receptor 1 complex, but also CYLD-independently. It will further determine how post-translational modifications of SPATA2 define a balance between TNF-induced survival, apoptosis, necroptosis and pyroptosis.
