Many tumor cells show resistance to the extrinsic and the intrinsic apoptosis pathways, yet their combined activation can override resistance and result in profound cell death induction. Using proteomics and structural mass spectrometry (MS), in particular crosslinking coupled to MS (XL-MS), this project will investigate how dynamic changes in the TRAIL receptor signaling complex and the BCL-2 interactome can sensitize hepatocellular carcinomas to the synergistic induction of apoptosis by TRAIL and chemotherapeutic drugs.
