TNF is a cytokine readily expressed and released under most inflammatory conditions. Intestinal epithelial cells are among the few cell types directly sensitive to TNF-induced cell death. This project will investigate how TNF-induced intestinal epithelial cell death is regulated by metabolites derived from the intestinal microbiome. It will further analyze how caspase 8-mediated cleavage of Gasdermins and associated pyroptosis contributes to TNF-induced cell death in intestinal stem cells versus mature epithelial cells.
