This proposal envisions a new platform for metabolomic classification in body fluids. At its core, this platform is based on compact high-field NMR (HyPERiON’s main objective), while the sensitivity is enhanced by parahydrogen hyperpolarization. This platform will operate on a three-step process: (1) preparation of a body fluid equivalent, (2) parahydrogen-based hyperpolarization and (3) sensitive NMR detection. While the platform itself is designed for generalized metabolite detection at low concentrations, special emphasis will be given to a target system. As target system, urine-based detection of analytes indicative of an infection with tuberculosis (TB) is chosen, a disease caused by the mycobacterium tuberculosis. We will prepare a laboratory version of the metabolomic cocktail with identical stoichiometry with the purpose to achieve detection sensitivity at the relevant molarity and chemical complexity.
