Studies from our laboratory have identified the mitochondrial 2-oxoglutarate dehydrogenase (OGDH) complex as a metabolic checkpoint whose activity determines whether endothelial cells (ECs) activate and divide or become quiescent. This function relies on the S(L)-enantiomer of 2-hydroxyglutarate (S-2HG), a metabolite inhibitor of several epigenetic enzymes, whose levels rise when OGDH activity is reduced. This project will study the role of this novel metabolic surveillance mechanism in adjusting endothelial responses during tissue damage, focussing on the interplay between metabolism and epigenetic gene regulation.
