The quorum-sensing receptors from Vibrio cholerae and Legionella pneumophila have six transmembrane ¿-helices, known ligands and regulate histidine kinases. The membrane anchors are identical in design as those of many adenylate cyclases. Ligand regulation by quorum-sensors was achieved in respective chimeric cyclases. We use bioinformatics, biochemical and physiological methods (cluster and structure analysis recombinant proteins, adenylate cyclase and ß-galaktosidase assays, MacConkey agar plates, FRET analysis) to study the mechanism of quorum-sensor transmembrane signaling. The data shall serve as a prelude to identify ligands of bacterial orphan receptors.
