Tissue-resident lymphocytes (TRL) can seed, persist and function in distinct tissue-environments, which they can modulate to facilitate local immune responses. In this project, we will establish engineering approaches for the generation of TRL-like cells for adoptive cell therapy. We will systematically interrogate and validate TRL-specific genes using CRISPR/Cas9-mediated screening. We will additionally explore the ability of engineered ILCs to modulate local environments through the targeted delivery of cytokines. The potential of these novel cell engineering strategies will be validated in experimental models of infection and tumor, and in human T cells.
