The multi-parameter analysis of cells within a heterogeneous sample at the single cell level is one of the most important technology for understanding of biological relationships and mechanisms in the context of health and disease. Key technologies are the analysis of single cells in suspension using flow cytometry or mass cytometry, as well as the analysis of cells in the tissue context using histopathology or imaging mass cytometry. Flow cytometry enables cells to be analyzed using a variety of parameters at high throughput. The high demand for highly specialized devices for both cell analysis and cell sorting and the high costs argue for a centralized provision of such technologies. The integration into a central core facility allows efficient use of the instruments and expert support for the users.The central Flow & Mass Cytometry Core Facility 'Charité | BIH Cytometry Core ' has been offering expert service and the latest equipment for multi-parameter single cell analysis and cell sorting at the Charité CVK site for more than 10 years. However, there is currently no centralized access to flow cytometry devices at the Charité Mitte site.The working groups at the Charité Mitte location involved in this application are dependent on the use of the equipment requested here for their interdisciplinary research projects. So far, devices from other institutes could be used for some of the projects or analyzes could be broken down into several approaches with fewer parameters. The devices of the other institutes are no longer available to the extent necessary. The limitation of the sample material increasingly requires a simultaneous analysis of as many parameters as possible.The Charité's central Flow & Mass Cytometry Core Facility therefore applies here for the purchase of an analytical 5-laser multi-parameter flow cytometer. Together with a 5-laser high-speed cell sorting device, this instrument should form the basis for the expansion of the central core facility also beyond the group of the applicants listed here. The integration of the instrument into the then cross-location central core facility under a common management structure should ensure the rapid availability of the technology. Established quality management structures are taken over and the training of the users is guaranteed through proven training concepts.
