Hepatocyte lipid metabolism does not only determine the lipid status of the liver itself, but is the major regulator of whole body lipid status and turnover with major implications in many types of disease. Due to experimental limitations in sensitivity and time resolution, our understanding of the dynamics of hepatic lipid metabolism is quite limited. As detailed below, we recently introduced new technology that offers strongly improved sensitivity and time resolution. This application describes further development of the technology and its use for better understanding the dynamics of hepatocyte lipid metabolism. The focus of the work will be on the two quantitatively major lipid classes, i.e. phosphatidylcholine and triacylglycerol.
