Chronic increased IL-6 levels in obesity lead to the development of hepatic IL-6 resistance where hepatocytes are unable to react to IL-6 signaling. Our data reveal that hepatic IL-6 resistance programs macrophages via hepatocyte-derived CSF-1 and IL-33, but the molecular consequences remain unknown. We have developed genetic mouse models to induce and prevent hepatic IL-6 resistance. Using these mice, we will investigate the impact of hepatic IL-6 resistance on macrophage programming with metabolic experiments and cell type-specific gene expression under dietary intervention.
