We aim to define the effects of local TH availability and efficacy in the liver in different cell compartments during progression of non-alcoholic fatty liver disease (NAFLD) to non-alcoholic steatohepatitis (NASH) and liver fibrosis. We will provide profound phenotypic, spatial and functional analyses including samples of a human cohort, iPSC derived cells as well as appropriate rodent models and a perfusable liver-on-a-chip system to improve our understanding of TH-dependent mechanisms driving or reversing metabolic injury, inflammation and fibrogenesis. Based on our findings we will be able to identify novel disease stage- and cell-type-dependent drug targets modulating TH actions.
