Hepatic TRß expression is reduced in human NASH, potentially causing local thyroid hormone (TH) resistance. This reduction was replicated in a novel NASH mouse model; however, NASH progression was lower in mice lacking TRß suggesting a protective effect. Moreover, our histological data showed that the lack of TRß affected the hepatic zones differently. We therefore aim in the next funding period to increase cellular resolution using single nuclei sequencing to decipher the role of TRß in fatty liver disease in our murine NASH model. Moreover, we will test the hypothesis that reducing TRß availability could be beneficial for NASH progression.
