This project aims to exploit modulation of local TH action for improved stroke outcome. In the first funding period we have shown that TH delivery in the acute phase of ischaemic stroke ameliorates brain damage and similarly that lack of endothelial TH transport in the blood-brain barrier is protective. We will now proceed to delineate spatiotemporal requirements and mode of TH action for improved stroke outcome in transgenic mice with cell-specific alterations in THT and TR signaling, and test long-term T3 treatment as well as T3-chimeras in ischaemic stroke models.
