Mass Spectrometry Platform (Part II): Ion Mobility Q-TOF Mass Spectrometer with Nano-HPLC System

The mass spectrometry platform described herein will be used for protein structure analysis in the recently established “Center for Structural Mass Spectrometry” (CSMS) headed by Prof. Sinz. The equipment will serve to conduct cooperative research projects with the Faculty of Natural Sciences I and the Medical Faculty at the Martin Luther University Halle-Wittenberg as well as within the RTG 2467 “Intrinsically Disordered Proteins – Molecular Principles, Cellular Functions, and Diseases” to investigate three-dimensional protein structures and to delineate protein-protein interactions. Both LC/MS systems have to fulfill all requirements to guarantee a robust, high-throughput analysis of proteins. There will be a clear separation of projects for the two ion mobility Q-TOF mass spectrometers in the CSMS: One LC/MS system will be employed for cross-linking/MS experiments, while the second LC/MS system will be used for conducting H/D exchange experiments. This will allow obtaining complementary data for the three-dimensional structural analysis of proteins and protein complexes up to system-wide protein network analyses. Establishing the CSMS will further strengthen the research focus on protein mass spectrometry at the MLU Halle-Wittenberg.
