We apply for a high resolution electrospray ionization tandem mass spectrometer coupled to nanoflow chromatography for proteome research at the University Medical Center Göttingen (UMG), University of Göttingen. The instrument we are applying for will be placed in the UMG's Scientific Core Facility Proteomics (DFG RIsources RI_00483) to offer access to this key technology in proteome research preferentially to the UMG's research groups. The Core Facility was founded in the UMG Institute of Clinical Chemistry in 2012, and is currently operating two high resolution mass spectrometers. The mass spectrometer applied for here will replace one of these aging instruments.We plan to mainly use the new instrument for the following applications:1) label-free proteome profiling in samples from cell culture, body fluids and tissues by data-independent acquisition (DIA) mass spectrometry2) global analysis of posttranslational protein modifications such as phosphorylation and ubiquitinylation after affinity enrichment3) analysis of protein interactions biomolecular complexes and cell compartments by affinity purification mass spectrometry or by proximity profiling methods4) analysis of direct protein/protein contact sites by chemical rosslinking using bifunctional crosslinking reagents
