This project is central to the Clinical Research Unit providing a platform for bioinformatics analysis and management of genomic and clinical data for all CRU 344 projects. Initial work on the CRU344 indicates the importance of interactions of myeloid cells, hematopoietic clones, and stromal cells in the progression of ET or PV to myelofibrosis. To tackle these questions, we will expand our bioinformatic pipelines, which are currently established for single cell and bulk transcriptomics analysis, to support analysis of clonal hematopoiesis via single cell sequencing and the analysis of cell-cell communications via spatial transcriptomics and proteomics. The methods will be used for the analysis of data generated by distinct projects of this CRU. Moreover, this service platform will create a clinical and genomic data warehouse to further improve our MPN clinical and genomic repositories. This data warehouse will allow standardization of rich and historical clinical data from the Aachen MPN bioregistry and the integrative analysis with the genomic data produced by this CRU. Moreover, we will expand genomic dataset by generating additional expression profiles of CD34+ positive cells of MPN patients and integration of sequencing data produced by all projects from this CRU.
