Cellular respiration is accomplished by a sophisticated machinery of membrane-bound, multiprotein and cofactor-bearing respiratory complexes that can form higher-order structures (supercomplexes). Assembly of these complexes is tightly regulated to avoid partially assembled malfunctioning complexes and to adapt the cell to changes in energetic demands. We will provide in-depth structural and mechanistic insight into the coordinated biogenesis of respiratory chain complex III and its related supercomplexes and also analyse the role of the mitochondrial signature lipid cardiolipin in this complex assembly process.
